The Welsh side have a played two and won two record and have a 100 percent record so far in the new expanded home and away tournament between sides from Ireland, Scotland and Wales.
The Lightning won their two opening games against Welsh rivals Brython Thunder during the festive period, while Clovers are fourth in the league after a win against reigning champions Wolfhounds in Round 2.
Welsh international Gwennan Hopkins captains the side, with Bryonie King sidelined with an ankle injury. Katherine Baverstock makes her first start in the Celtic Challenge after impressing off the bench.
Wales international Carys Williams-Morris is selected at centre for the injured Kelsie Webster and partners Molly Anderson-Thomas in the Lightning midfield.
Lottie Buffery-Latham and Nia Grundy are both named on the Gwalia bench and will make their first bow in the league.
