The Welsh side return home looking for a first win of the cross-border league against the second in the table Irish side at the home of the Scarlets.
Welsh international Natalia John captains the side for a fourth successive game and is partnered by Wales U20s lock Robyn Davies in the second row.
Brython Thunder have made four changes with one positional to the side that narrowly went down to league leaders Edinburgh, with Davies moving to lock from blindside flanker
Wales centre Meg Webb, hooker Poppy Hughes, and flanker Kira Philpott all come into the starting line-up.
Webb will form an all-Wales centre partnership with fellow Test star Hannah Bluck, who scored a brace of tries against Edinburgh, in the Thunder midfield.
Wales second row Gwen Crabb has been recalled by her club and was not considered for selection.
Brython Thunder’s performances have improved in every game during the campaign but they face a Wolfounds side full of confidence with two wins in the competition.
Ashley Beck, Head coach of Brython Thunder, said: “This is a new-look Brython Thunder squad this season and we have improved in every game we have played so far in the Celtic Challenge.
“We produced another good performance against Edinburgh and were unlucky not to get more out of that game but the character of the side really came through in the Scottish capital.
“We need to build on that performance but know the Wolfhounds will be looking to win and will have their eyes on top spot in the table. We are very aware how strong the Irish sides were in last season’s Celtic Challenge.”
