The Welsh side travel to Ireland on the back of a three-match winning streak to face the reigning champions in their own backyard.
Welsh international Bryonie King captains the side which sees two changes from the starting line-up that did the double over Edinburgh.
King is selected in a backrow including fellow Wales Test star Gwennan Hopkins and Catrin Stewart.
The Gwalia pack sees one change with Danyelle Dinapoli coming into the front row, alongside Maisie Davies and Molly Wakely.
Welsh international Alaw Pyrs partners Lily Terry in the Lightning engine room.
Wales full back Jenny Hesketh, who came off the bench in the win over Edinburgh, makes her first start for Gwalia Lightning.
Fellow Welsh international Caitlin Lewis and Courtney Greenway are selected on the wings and join Hesketh in the Lightning back three.
The centre partnership of Kelsie Webster and Anwen Owen, who impressed against Edinburgh are selected in midfield, with fly half Carys Hughes and Wales scrum half Sian Jones at half back.
