Eddie Jones’ team arrive in the Welsh capital this weekend with both outfits level on four points following victories over France and Italy respectively on the opening weekend.
The Red Rose brigade are unbeaten in their last 15 matches under Australian head coach Jones, with Wales aware of the huge threat the men in white will pose.
Howley has made two changes to his side for the encounter – replacing props Nicky Smith and Samson Lee with Rob Evans and Tomas Francis.
The former scrum half said: “England have gone 15 games unbeaten, they’ll have huge confidence and self-belief. They got across the line against France and there’s no doubt they’ll be ready for this game. We’re looking forward to kick-off on Saturday.
“Games between Wales and England are always tight affairs. It’s a game the players, supporters and coaches look forward to. It’s another challenge for us to embrace.”
Saturday’s game in the Welsh capital will see the Principality Stadium roof open, at the request of England boss Jones. And Howley, who has named Dan Biggar and George North in his starting XV, is looking forward to the challenge.
“It’s about the mental aspect going into the game – with the six-day turnaround,” he added.
“We’ve tried our best to prepare as a side. The boys did cryotherapy on Sunday evening. Those two players (Biggar and North) will be given every opportunity to prove their fitness.
“They’re important players and both want to be part of that team, as you’d expect. We’ve got extremely motivated players and are looking forward to the challenge England will bring.”
Wales will be led once again by lock Alun Wyn Jones, who has a perfect record from his three Six Nations games as captain of his country.
“We’re fortunate to have a stadium that adds to the occasion, but the rugby has to be the focus and hopefully we can do the occasion justice. It’s up to us to show that on the pitch,” Jones said.
“This is the sort of occasion you want to be involved in.”
