The champions are one of five Premiership clubs who find themselves playing against Championship opposition in the first round and they will get an early chance to see how strong their opponents will be when they meet in a pre-season friendly on 22 August.
Premiership winners Pontypridd, who were beaten at the Millennium Stadium in the Cup final, have to travel to Swansea, while Bedwas will also be on the road against a Championship club when they go to Pontypool.
New Neath forwards coach Gareth Llewellyn will have to find a way to tame the best pack in the Championship last season when Bargoed go to The Gnoll. RGC1404 were Cup winners in Neath last season and they host Llanharan in the first round.
