Keys were the beaten finalists in 2014, won the cup for the first time in 2012 and are currently second in the Principality Premiership.
RGC 1404’s reward for their 15 try, 93-0 home win over Llanharan last weekend is another home tie, this time against Premiership opposition. Aberavon, who received a bye in the first round, will make the long trek north.
Last season RGC 1404 managed to beat Neath in the first round before falling to another Premiership side, Cross Keys, at home in the second round.
There are three Premiership sides who will definitely be at home in round two with Llandovery entertain Newbridge following their victory last weekend over Cardiff, Llanelli will play the winners of the tie between Narberth and Merthyr and Pontypridd will host Bridgend Athletic.
