The league leaders are without Wales stars Alun Wyn Jones, Dan Biggar, Rhys Webb and Scott Baldwin who are in the starting lineup to take on the Springboks this weekend.
James King and Aaron Jarvis are also unavailable after being named on the bench, but Justin Tipuric could return to regional action in Dublin. Leinster are unbeaten in five matches since their PRO12 defeat to rivals Munster at the Aviva Stadium.
But there is hope for the Ospreys, who are the last team to take points off Leinster at the RDS after their 29-all draw in September 2013.
The two sides met on four occasions last season with Leinster taking both encounters in the European Cup but Ospreys securing a win and a draw in the PRO12. Ospreys have played nine previous PRO12 matches at the RDS Arena, winning three, drawing one and losing five.
Fly-half Sam Davies said: “The boys showed some good grit and determination to come back from 18-3 down and we were gutted not to come away with a win, let alone a bonus-point. But there is a lot to build on, if we can sure up our line-out and scrum and get better set-piece ball, then we can have a real crack at it.
“I enjoy Martin Roberts’ service and chat. It’s been a great experience to play with someone like him and hopefully we can build that relationship against Leinster and give it a real good go.
“You have to take the games as they come, we can’t change anything now. I’m confident we can beat them if we can get the ball for long periods especially with our backline and the skills we’ve got.”
